Propagation of Hantaan virus in Mongolian Gerbil Fibroblasts and Utility of MGF for Immunofluorescence Assay Antigen Slide



Abstract
Mongolian gerbil fibroblasts(MGF) derived from Mongolian gerbil embryo were established by primary culture and Hantaan virus was propagated in those cells. MGF were more rapidly infected with Hantaan virus than VeroE6 cells which were ordinarily
used
for propagation of Hantaan virus. We used Hantaan virus-infected MGF as an antigen for indirect immunofluorescence assay for the diagnosis of hemorrhagic fever with renal syndrome. In comparison with Hantaan virus-infected VeroE6 cells, Hantaan
virus
infected MGF were more sensitive to detect viral antibodies and the specificity was equal. But the titer of Hantaan virus released to culture medium from MGF was 10 fold lower than that from VeroE6 cells.
Compared to VeroE6 cell, MGF have larger cytoplasmic compartment, where Hantaan viral proteins are mainly expressed in, thus show more distinct immunofluorescence pattern of the viral proteins. These results indicate that the interpretation of
immunofluorescence assay in MGF was easy and clear. We suggest that immunofluorescence assay using Hantaan virus-infected MGF is useful in diagnosis of Hantaan virus infection.
